Sterile Procedure and Practice
***Always be sure to wipe down your area and tools with 70% iso or a 10 % bleach solution, 70% iso is recommended as it is better for killing bacteria and mold spores then 99% iso. 70% iso contains 30 % water which breaks down cell walls and allows the iso to penetrate and kill the bacteria mold or organism.
***When working with cultures or doing transfers of any kind it is best to eliminate any airflow or movement beside that of your laminar flow hood as this unclean air may contain contaminants especially when using a still air box. It is also recommended you run your flow hood for 30minutes to 1 hour before use this will scrub the air of any particulate contaminate
***When using a SAB Still air box it is best to eliminate any air currents within the area you are working an hour prior to use to help combat contaminates in the air. Also it is best to make slow calculated movements to prevent air currents within the still air box. Quick movements may cause a vacuum of air which can pull contaminated air into the SAB and cause contamination. Sterilize your SAB with 10 % bleach solution or 70% ISO.
***I would also recommend having a shower prior to doing any culture work. Wearing a mask to prevent bacteria from your breath. Using latex powder free gloves and a change of freshly laundered clothing as this will also eliminate contamination.
*** Ideally you want to be doing this all in a very clean environment with very little organic materials, anything untreated that has many crevices or pores will hold or contain contaminates harmful to your mycelia.
